Pengaruh Pemberian Recombinant Human Erytropoietin (RHuEpo) Terhadap Ekspresi Protein Bax Pada Neuron Hipokampus Tikus (Rattus Norvegicus) Pasca Ligasi Arteri Carotis Communis Unilateral

Abstract


Oklusi pembuluh darah otak dapat menginduksi stroke iskemik. Hippocampus rentan terhadap hipoksia. Saat ini, terapi stroke iskemik terfokus pada penghilangan oklusi. Pengembangan agen neuroprotektif Recombinant Human erythropoietin (RHuEpo) untuk mencegah kematian sel otak masih dibutuhkan. Salah satu biomarker apoptosis adalah protein Bax. Terhambatnya kematian sel dapat dibuktikan dengan penurunan Bax. Tujuan penelitian. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian Recombinant Human erythropoietin (RHuEpo) terhadap ekspresi protein Bax pada tikus pasca ligasi arteri karotis komunis unilateral (UCCAO). Metode. Rancangan penelitian adalah eksperimental post test only control group. Subjek penelitian adalah 24 ekor tikus wistar jantan, terbagi menjadi 4 kelompok. Kelompok Sham (S) adalah kelompok tikus yang tidak mendapatkan tindakan UCCAO. Kelompok 1, 2 dan 3 adalah kelompok perlakuan P1 (induksi UCCAO), kelompok perlakuan P2 (UCCAO + RHuEpo 400 IU/hari), dan kelompok perlakuan P3 (UCCAO + RHuEpo 800 IU/hari). Ekspresi Bax neuron Hippocampus diamati pada sediaan preparat dengan pewarnaan imunohistokimian. Uji analisis menggunakan SPSS One Way Annova. Hasil. Terdapat perbedaan bermakna ekspresi Bax neuron hippocampus antara kelompok P1 dengan P3 (p=0.029). Jumlah ekspresi Bax neuron kelompok P1 lebih tinggi dari P3. Kesimpulan. Terdapat pengaruh pemberian Recombinant Human erythropoietin (RHuEpo) terhadap ekspresi Bax hippocampus tikus yang diinduksi UCCAO.
